Abstract

The plant utilized in this study is the Nampu leaf (Homalomena latifrons Engi.), which belongs to the Araceae family. The research aims to determine the flavonoid content in the extract of Nampu leaves (H. latifrons Engi.) based on solvent variations. The extraction of Nampu leaf plants (H. latifrons Engi.) was carried out using a multi-step maceration method with solvents such as 70% ethanol, methanol, and distilled water. Qualitative analysis was performed using thin-layer chromatography (TLC) characterized by the appearance of fluorescent spots on the TLC plate under a UV lamp at 366 nm, with positive spraying of AlCl3 forming yellow-greenish spots indicating the presence of flavonoid compounds. Subsequently, the determination of the total flavonoid content was conducted using a UV-Vis spectrophotometer method employing quercetin standards measured at a maximum wavelength of 431 nm. The research findings indicate that the total flavonoid content in the Nampu leaf extract (H. latifrons Engi.) is 27.69 mgQE/g extract or 2.76% (w/w) for 70% ethanol extract, 58.29 mgQE/g extract or 5.82 % (w/w) for methanol extract, and 12.12 mgQE/g extract or 1.21% (w/w) for water extract. The most optimal solvent for seeking flavonoid compound content in Nampu leaf plants (H. latifrons Engi.) is methanol.
